Injectin In Vivo siRNA Delivery Kit contains a new lipid-based formulation which enables the delivery of siRNA into living organisms. Injectin reagent has been especially developed for local and systemic siRNA administrations in mouse. Due to it's high efficiency and it's stability, Injectin reagent is able to maintain in vivo silencing of gene expression for a very long time, without the need of multiple injection repeats.

The siRNA delivery protocol given for the delivery of 10 µg of siRNA into a mouse tumor. A table is provided to adapt your protocol for other siRNA delivery routes.
    1. Dilute 10 µg of siRNA into 10 µL of glucose containing buffer.
    2. Complete to 40 µL with a sterile RNase-free water.
    3. Add 10 µL of Injectin reagent and mix by pipetting up and down.
    4. Incubate 15 minutes at room temperature.
    5. Inject in mouse tumor.
    6. Monitor gene expression minimum 72 h after injection.

  • Storage and Handling
Upon receipt, the kit should be stored at 4°C. Stored properly, the kit components should remain stable for at least 1 year.
Figure 1: 10 µg of a luciferase siRNA were directly injected into a 100 mm3 mouse tumor 4T1 expressing luciferase by using 10 µL of Injectin reagent. The same experiment was performed by using other in vivo siRNA delivery reagents (X, Y and Z) according to manufacturer's protocols. After 8 days, tumors were excised and lysed in a protease inhibitor solution. Luciferase activities were measured with a luminometer. All data were corrected by protein concentration assays.
