.TALENs are engineered DNA-cleaving enzymes made by fusing tailor-made DNA-binding peptidic modules to the FokI nuclease domain. TALENs induce site-specific double strand breaks (DSBs) in DNA at user-defined locations, which leads to targeted modification of the genome. Each TALE module recognizes a single nucleotide base. The fusion of 15-20 modules gives an unsurpassed sequence specificity to each TALE, and the use of a TALE pair is then considerably accurate. Recent publications have established TALENs as an innovative tool for targeted genome editing in diverse cell lines and organisms (method of the year 2011 - Nature Methods Jan 2012). Because the DNA-binding domain can be designed to recognize and introduce DNA double-strand breaks specifically in desired sequences in the genome, TALENs provide an innovative tool for in vivo genome editing. Using our custom-synthesized TALENs, scientists can knock out any gene of interest or edit its DNA sequence in cultured cells or other model systems such as zebrafish, mouse, rat, plants, etc.

TALENs make various types of genome editing possible, including, but not limited to, (1) the introduction of mutations into a gene of interest to knockout its function, (2) the deletion or insertion of genetic information at specific loci, and (3) the induction of large scale genetic deletions, duplications, and inversions. These types of genome editing were difficult, if not impossible, to achieve before ZFNs and TALENs were developed; the ability to undertake such editing represents a unique opportunity in both basic and pharmaceutical research to translate our growing understanding of the genetic basis of many biological and pathological processes into industrial and therapeutic breakthroughs.
