- AccuTargetTM Positive siRNAs can induce effective knockdown of target genes. As an endogenous gene, we offer a positive control siRNA targeting GAPDH and as a reporter system, positive control siRNAs targeting GFP and luciferase genes are available.
Hela cells were transfected with GAPDH- and NC(negative control)-siRNA at 100 nM final concentration using lipofectamine 2000 reagent. Twenty four hours post-transfection, total cellular RNA was isolated from transfected cells and subjected to Northern blot and real-time PCR analyses. As can be seen in Fig. 12B, only about 3% GAPDH mRNA remained after 24 h treatment with GAPDH siRNA, indicating that highly efficient knockdown of GAPDH mRNA can be easily achieved using our positive control GAPDH siRNA.
