.RNA-Guided ENdonucleases (RGENs) are novel, programmable genome engineering tools that were developed from bacterial adaptive immune machinery that serve to identify and destroy foreign DNA. In this system — the clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated (Cas) immune response — the protein Cas9 forms a sequence-specific endonuclease when complexed with two RNAs, one of which guides target selection. Likewise, RGENs consist of constant components (Cas9 and tracrRNA) and a target-specific RNA (crRNA).

RGENs utilize a short guide RNA (gRNA) to recognize DNA, bind the Cas9 endonuclease, and induce site specific cleavage. The gRNA directs Cas9 to the target sequence where it unwinds and cleaves the double stranded DNA.

Thus, unlike current genome engineering tools, such as zinc finger nucleases (ZFNs) and TAL effector nucleases (TALENs), RGENs use the unambiguous interaction between guide RNA and target DNA for target site selection, which is expected to provide superior specificity. As such, RGENs represent a new generation of engineered nucleases.

RGENs recognize a target sequence that is 23bp in length and ends with two guanines (GG). This minimal requirement for target site selection means that RGENs can be designed to cleave any region of the genome (every 8bp, in theory). Specific digestion of your target sequence in the cell allows efficient genome engineering (knockout / knockin / locus deletion) in vivo.

RGENs have recently been shown to support efficient genome engineering in human cells, zebrafish and bacteria ( Cho et al. 2013. Nature Biotech ., Cong et al. 2013. Science , Hwang et al. 2013 Nature Biotech ., Jiang et al. 2013 Nature Biotech ., Mali et al. 2013. Science ). In addition, the use of RGENs allows multiplexing of genome engineering processes (e.g., knockout of multiple genes in a cell at one time). RGEN applications will expand to various plant and animal cells/organisms soon.
